No you can play alone and it will unlock as soon as you finish the level, tested on an alt account

Did you play with inf health on?does it work?I've tried it on Ninja Gaiden 3 Razors Edge but eventually Health gets low when I I've got hit too many times?

No, no cheats at all were used. Just play the easiest mission in team mission mode, you shouldn't die at all if you know basic combat
